چکیده
Abstract BACKGROUND Glioblastoma multiforme (GBM) remains a disease with debilitating survival outcomes. Cancer immunogenomics represents complementary approach to the application of genomics in developing novel immunotherapies for brain malignancies. Using cancer that we developed, called Open Reading Frame Antigen Network (O.R.A.N.), identified immunogenic neoantigens and tumor-associated antigens (TAAs) including cancer-testis developmental antigens, are aberrantly overexpressed murine models GBM. The aim this study is evaluate immunologic effects safety immunotherapy vaccines targeting TAAs preclinical METHODS RNAseq WES were performed glioblastoma tumors- KR158-Luc GL261, results fed O.R.A.N pipeline antigen prediction. A tumor antigen-specific RNA library was created each using gene enrichment strategy validated by RNAseq. Tumor-bearing animals treated adoptively transferred ex vivo expanded lymphocytes dendritic cell loaded RNA. Tumor volume, thus progress, determined luciferase imaging technique outcomes noted. We also evaluated efficacy combination checkpoint blockade therapy treating tumor-bearing combined an anti-PD-1 antibody. RESULTS significantly effective slowing progression GBM tumors both adoptive cellular as well therapy. Additionally, T cells several our predicted increase tumor-infiltrating memory animals. CONCLUSION developed cell-based vaccination being tumor-specific

Immunotherapy for the treatment of glioblastoma.
Glioblastoma, the most aggressive primary brain tumor, thrives in a microenvironment of relative immunosuppression within the relatively immune-privileged central nervous system. Despite treatments with surgery, radiation therapy, and chemotherapy, prognosis remains poor. The recent success of immunotherapy in the treatment of other cancers has renewed interest in vaccine therapy for the treatm...
متن کاملImmunotherapy for Breast Cancer Treatment
Breast cancer, as a heterogeneous disease, includes a wide range of pathological and clinical behaviors. Current treatment protocols, including radiotherapy, chemotherapy, and hormone replacement therapy, are mainly associated with poor response and high rate of recurrence. Therefore, more efforts are needed to develop alternative therapies for this type of cancer. Immunotherapy, as a novel str...
متن کاملImmunotherapy for Glioblastoma
Glioblastoma (GBM) is the most common primary malignant brain cancer with a dismal prognosis in spite of aggressive treatment options. Although once thought to be an “immune-privileged” site, recent advances have begun to highlight the complex interaction between the immune system and the central nervous system. Thus, great interest has emerged in the ability of immunotherapy to potentially pro...
متن کاملVirosome vaccines and their application in cancer immunotherapy
Background: An increasing number of tumor associated antigens (TAA) capable of inducing immune responses have been identified in the last two decades. Unfortunately, they are weak immunogens and require potent adjuvants to promote their immunogenicity. Virosomes, nano-lipid vesicles containing viral protein spikes, have been proven amenable for transferring antigen (Ag) to the major histocompat...
متن کاملDendritic Cell-Based Immunotherapy Treatment for Glioblastoma Multiforme
Glioblastoma multiforme (GBM) is the most malignant glioma and patients diagnosed with this disease had poor outcomes even treated with the combination of conventional treatment (surgery, chemotherapy, and radiation). Dendritic cells (DCs) are the most powerful antigen presenting cells and DC-based vaccination has the potential to target and eliminate GBM cells and enhance the responses of thes...
